The one-way journey
Unusually practical for a linear trail of this length: unlike most of the other trails in this dataset, the South Downs Way has three intermediate stations (Amberley, Southease and Liss) that the National Park Authority itself names as genuine day-walk start/end points, plus Lewes as a bigger nearby hub — making section-walking over several separate trips, rather than one continuous through-walk, an unusually normal way to do this trail. For a single through-walk, most people simply train to Winchester and train home from Eastbourne without needing a car at all.
If you need to leave the trail early
The intermediate stations (Amberley, Southease, Liss, plus Lewes nearby) are the single most reliable bail-out option, being directly on or very close to the path and served year-round. The bus picture is patchier and more day-specific than it first appears: the Breeze 77/79 buses reach Devil's Dyke and Ditchling Beacon from Brighton but their exact season wasn't confirmed here; the Coaster 12 covers the Seaford-Eastbourne stretch; and the Cuckmere Buses network around Alfriston, while genuinely useful, is a patchwork of once- or twice-a-week community services (different routes running on different specific days, e.g. Wed/Fri, Tue/Thu/Sat, Sundays only) rather than a daily service — check the exact day before planning around it.

About 77km into the trail. The South Downs National Park Authority's own site names Amberley as one of only three stations (with Southease and Liss) you can begin or end a day's walk from directly — the station sits right by the Arun Valley crossing on the route.

About 123km into the trail — not directly on the route but the largest town near it in the eastern third, and a genuine transport hub (Southern services towards Brighton, Eastbourne and London via Haywards Heath/Gatwick). About 134km into the trail. The South Downs Way literally crosses the Seaford branch line here, per Wikipedia's own description of the station — about as direct a trail/rail intersection as exists on any trail in this dataset, and one of the three stations (with Amberley and Liss) the National Park Authority itself names as a genuine start/end point for a day's walk.
